Did you test for growth promotion your media (I guess it was NIH) ? Which
strain did you use (only anaerobic or aerobic also) ?

I am wondering about the condition of the media in those vials: is NIH
itself able to keep anaerobic conditions in media filled vials ?

I have performed them once using Fluid Thio for a client.  If you choose to
use this media be sure that you order it without agar.  However
soybean-casein-medium should be sufficient.
