David,
We use the graduations on the funnel to measure the volume, and have
validated this method  by measuring sample weight.
The vacuum is switched off until the correct volume has been measured out
and then vacuum applied.  If the vacuum is on while measuring a larger
volume than required will be dispensed.

I would like to elicit the help of the PMF group to clarify an internal
discussion! When sampling 100-200mL of sample e.g. water, into a
disposable filter funnel for filtration do you use the volume marks
printed on the funnel side to determine volume or pre-measure the sample
volume in a separate container. If the former do you have a spec for
volume as >100mL or something else. Secondly when preparing the sample,
is the filtration manifold vacuum turned on while pouring the sample
into the funnel or is the volume poured then the vacuum applied to pull
the sample through?"
