If you use the mark on the funnel you need to verify the accuracy of
that mark, set a frequency, e.g., one per lot.  What many do for
drinking water is to use a sterile graduation cylinder (buy high quality
glassware and verify one per lot).

Turn on filtration vacuum after the sample has been added otherwise the
sample/bacteria will concentrate at the spot where you just poured in.

I would like to elicit the help of the PMF group to clarify an internal
discussion! When sampling 100-200mL of sample e.g. water, into a
disposable filter funnel for filtration do you use the volume marks
printed on the funnel side to determine volume or pre-measure the sample
volume in a separate container. If the former do you have a spec for
volume as >100mL or something else. Secondly when preparing the sample,
is the filtration manifold vacuum turned on while pouring the sample
into the funnel or is the volume poured then the vacuum applied to pull
the sample through?"
