Dear Prakash,

At first, as per the regulatory standards, any National level regulated
culture collection centres are valid and acceptable to receive the standard
cultures for industrial use (like GPT of media and validation studies) e.g.
MTCC, NCL etc. in INDIA.

Secondly better to store the cultures using Cryopreservation technique
(storing the prepared stock cryo cultures at about -80 C for infinitve time
as per ATCC literatures) then the conventional slant technique. Even you can
get the working cultures of as low as 3rd passage as per the procedure set
forth, for the said purpose. (NOTE: It is vital to use the lowest possible
passage in testing to get the best microbial response). However,
for your procedure it can be demonstrated through a validation study for the
storing of slant cultures.

You need to consider few things for this validation, like drying of agar
slants, viability of the stored cultures, and of course the storage time
period (in general 1 to 3 months is demonstratable; you have to give
transfer of cultures to fresh slants every 1 to 3 months). beyond six months
of storage of slant medium it was seen that the slants got dried.
